Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
After injection, the peptide binds receptors on somatotroph cells in the anterior pituitary. Receptor activation raises intracellular cyclic AMP and triggers release of stored growth hormone into the bloodstream. Because the compound works through the body's own regulatory system, growth hormone pulses retain much of their normal feedback control. Repeated administration also raises insulin-like growth factor 1, a hormone produced mainly in the liver. Investigators treat that rise as a marker that the pituitary axis has been engaged.
Clinical study of tesamorelin has centered on adults with HIV-associated lipodystrophy, a condition in which abdominal fat accumulates while peripheral fat is lost. In controlled trials, treated participants showed reductions in visceral adipose tissue measured by imaging, alongside modest shifts in some lipid values. Effects on subcutaneous fat were smaller and less consistent across studies. Whether these changes translate into fewer cardiovascular events remains an open question, because the trials were not designed or powered to answer it.
Regulatory approval in the United States came in 2010, when the Food and Drug Administration cleared the peptide for the reduction of excess abdominal fat in adults with HIV infection and associated lipodystrophy. The decision rested mainly on two randomized phase 3 trials that enrolled roughly eight hundred patients and ran for twenty-six weeks. Participants receiving active drug showed substantially greater declines in visceral adipose tissue than those receiving placebo, while total body weight changed comparatively little. A reformulated presentation was later approved, and the product has remained a niche therapy rather than a general weight-loss agent.
Tesamorelin occupies a narrow position among agents that act on the growth hormone axis. Unlike growth hormone itself, which is given as replacement, it stimulates the pituitary to release the hormone in pulses, so the downstream increase in insulin-like growth factor 1 depends on intact somatotroph function. Other peptides in the same family include shorter GHRH fragments and synthetic secretagogues with different stability profiles. Several points remain unresolved, including whether the reduction in visceral fat translates into fewer cardiovascular events, what happens to metabolic markers after long-term use, and how the drug compares with lifestyle or surgical approaches.

A documented effect of tesamorelin is a reduction in visceral adipose tissue in some study populations. Researchers have reported decreases in trunk fat measured by computed tomography alongside changes in lipid markers. The mechanism is thought to involve growth hormone-mediated lipolysis, though the precise contribution of direct versus indirect pathways is not fully resolved. Studies have generally examined defined groups over finite periods, so long-term outcomes are less well characterized. Findings have not been uniform across all trials.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone (GHRH). Its sequence corresponds to the 44-amino-acid form of human GHRH with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification slows enzymatic cleavage and extends the peptide's activity relative to the native hormone. The compound is produced by solid-phase peptide synthesis and supplied as a lyophilized powder. Researchers classify it as a GHRH receptor agonist. Its structure places it in the same family as other growth hormone secretagogues that act on the pituitary.
Binding of tesamorelin to GHRH receptors on pituitary somatotroph cells triggers cyclic AMP signaling and the release of growth hormone into circulation. Because the peptide acts upstream of the growth hormone axis, its effects are partly mediated by hepatic insulin-like growth factor 1 (IGF-1) production. The pulsatile character of endogenous growth hormone secretion is preserved rather than replaced. Whether amplified signaling produces effects beyond those of native GHRH remains an area of ongoing investigation.
Several compounds share the GHRH framework, including sermorelin, the shorter 1-29 fragment, and other analogs built on the full 1-44 chain. Naming follows a common convention: a stem that identifies the peptide plus a suffix marking analog status. Reports may describe tesamorelin by its sequence fragment, as a GHRH(1-44) analog, or by its amino-terminal modification. Indexing the compound therefore requires searching all of these forms, since some older literature predates the current international nonproprietary name.
Tesamorelin is a synthetic peptide built from 44 amino acids and classified with the growth hormone–releasing hormone family. Its sequence corresponds to the human GHRH(1-44) backbone, carrying one structural change at the amino terminus. That change is a trans-3-hexenoyl group placed where the natural peptide would have an unmodified end. The modification is the feature that separates the compound from the endogenous hormone in name, in stability, and in how it is handled in the laboratory.
== History == In 1992, Professor John T. Isaacs of The Johns Hopkins School of Medicine in Baltimore, Maryland, was investigating ways to approach the treatment of slow-growing tumors and discovered they could be efficiently destroyed if intracellular calcium levels were raised significantly. He determined that the ideal agent for upsetting intracellular calcium levels was the novel chemical thapsigargin. He began collaborating with Dr. Soren Christensen, a natural products chemist, at the University of Copenhagen in Denmark, and the first scientist to isolate thapsigargin from the plant Thapsia garganica, a poisonous weed that grows wild in areas of the Mediterranean. The objective of the collaboration was to chemically alter thapsigargin to a derivative that could be linked to peptides, thus forming prodrugs to treat certain tumors. Drs. Isaacs and Christensen were joined in their research by Dr. Hans Lilja, now at the Memorial Sloan-Kettering Cancer Center in New York, and Dr. Samuel Denmeade, of the Johns Hopkins School of Medicine. Together the four co-inventors developed a technology platform in which the therapeutic component of all the compounds would remain the same, but the targeting peptide would change. Peptides would be selected that could only be "cut off" or cleaved by enzymes specific for certain tumors. In March 2016, CEO and CFO, Craig A. Dionne resigned and Peter E. Grebow was announced as interim CEO. On August 1, 2016 the company changed its name to Inspyr Therapeutics and began trading under NSPX.
==== Caesarean section ==== Women are less likely to start breastfeeding after caesarean delivery compared with vaginal delivery. Though the stress of a caesarean section (c-section) may impact let-down and flow, and though time for recovery immediately after surgery may be needed, women who give birth via c-section are still able to breastfeed. Generally, the benefits of breastfeeding outweigh the harms posed by pain medications following surgery. Mothers may need to experiment with different breastfeeding positions to avoid putting pressure on the incision.

==== Canada, United Kingdom, and Europe ==== In Canada, e-cigarettes had an estimated value of 140 million CAD in 2015. There are numerous e-cigarette retail shops in Canada. A 2014 audit of retailers in four Canadian cities found that 94% of grocery stores, convenience stores, and tobacconist shops which sold e-cigarettes sold nicotine-free varieties only, while all vape shops stocked at least one nicotine-containing product. By 2015, the e-cigarette market had only reached a twentieth of the size of the tobacco market in the UK. In the UK in 2015 the "most prominent brands of cigalikes" were owned by tobacco companies, however, with the exception of one model, all the tank types came from "non-tobacco industry companies". Yet some tobacco industry products, while using prefilled cartridges, resemble tank models. By 2023, 1 survey by the Local Data Company counted 3,573 specialist vape shops in the UK, and NIQ data cited in press reporting put value sales of vaping products in Britain at £897.4 million. Sky News similarly reported that more than 230 independent vape shops opened in 2023, citing the same Local Data Company survey. France's e-cigarette market was estimated by Groupe Xerfi to be €130 million in 2015. Additionally, France's e-liquid market was estimated at €265 million. In December 2015, there were 2,400 vape shops in France, 400 fewer than in March of the same year. Industry organization Fivape said the reduction was due to consolidation, not to reduced demand.
=== Africa === Lasopy soucril (French: zizi de zebu) is a zebu penis soup in Malagasy cuisine. It is seen as an aphrodisiac and a treatment for infertility and erectile dysfunction. The penis is soaked and par-boiled to remove any remaining urine, then boiled with vegetables for several hours. Lasopy soucril is served at a high temperature with pepper and chilies.
=== Pricing and Profits === Throughout B.P.C. control, significant profits were made. In 1948, revenues from the island's phosphate reached $745,000. As the B.P.C. was controlled by its partner governments and was a major supplier of phosphate, it had an effective monopoly over the supply of phosphate to the markets in Australia and New Zealand, and could determine the price of phosphate delivered to those markets. The B.P.C. tied the pice for Ocean Island phosphate to the price paid for Nauru phosphate, however, the BPC would vary the price paid for phosphate. For example, around 1970, Nauru phosphate was sold to Japan at $14 and $15 a ton, but sold into the markets in Australia and New Zealand at $12.30.
==== United States ==== 2C-EF is not an explicitly controlled substance in the United States. However, it could be considered a controlled substance under the Federal Analogue Act if intended for human consumption.
Many mothers have to return to work a short time after their babies have been born. In the U.S., about 70% of mothers with children younger than three years old work full-time, with 1/3 of the mothers returning to work within 3 months and 2/3 returning within 6 months. Working outside of the home and full-time work are significantly associated with lower rates of breastfeeding and breastfeeding for a shorter duration of time. According to the Centers for Disease Control and Prevention, support for breastfeeding in the workplace includes several types of employee benefits and services, including writing corporate policies to support breastfeeding women; teaching employees about breastfeeding; providing designated private space for breastfeeding or expressing milk; allowing flextime to support milk expression during work; giving mothers options for returning to work, such as remote work, part-time jobs, and extended maternity leave; providing on-site or near-site child care; providing high-quality breast pumps; and offering professional lactation consultants. Programs to promote and assist nursing mothers have been found to help maintain breastfeeding. In the United States the CDC reports on a study that "examined the effect of corporate lactation programs on breastfeeding behavior among employed women in California [which] included prenatal classes, perinatal counseling, and lactation management after the return to work".

Ion yield is typically estimated to range from 10−4 to 10−7, with some experiments hinting to even lower yields of 10−9. The issue of low ion yields had been addressed, already shortly after introduction of MALDI by various attempts, including post-ionization utilizing a second laser. Most of these attempts showed only limited success, with low signal increases. This might be attributed to the fact that axial time-of-flight instruments were used, which operate at pressures in the source region of 10−5 to 10−6, which results in rapid plume expansion with particle velocities of up to 1000 m/s. In 2015, successful laser post-ionization was reported, using a modified MALDI source operated at an elevated pressure of ~3 mbar coupled to an orthogonal time-of-flight mass analyzer, and employing a wavelength-tunable post-ionization laser, operated at wavelength from 260 nm to 280 nm, below the two-photon ionization threshold of the matrices used, which elevated ion yields of several lipids and small molecules by up to three orders of magnitude. This approach, called MALDI-2, due to the second laser, and the second MALDI-like ionization process, was afterwards adopted for other mass spectrometers, all equipped with sources operating in the low mbar range.
== Biosynthesis and industrial production == Tryptophan is not synthesized from simpler substances in humans and other animals and is sourced from dietary intake of tryptophan-containing proteins. Plants and microorganisms commonly synthesize tryptophan from shikimic acid or anthranilate. Tryptophan biosynthesis starts with the condensation of anthranilate with phosphoribosylpyrophosphate (PRPP), generating pyrophosphate as a by-product. The ring of the ribose moiety is opened and subjected to reductive decarboxylation, producing indole-3-glycerol phosphate; this, in turn, is transformed into indole. In the last step, tryptophan synthase catalyzes the formation of tryptophan from indole and the amino acid serine.
However, benzodiazepines have been used clinically to manage the adverse psychological effects of psychedelics, for instance in clinical studies and in the emergency department. A clinical trial of psilocybin and midazolam coadministration found that midazolam clouded the effects of psilocybin and impaired memory of the experience. Benzodiazepines might interfere with the therapeutic effects of psychedelics, such as sustained antidepressant effects. Some serotonergic psychedelics, for instance dimethyltryptamine (DMT) and 5-MeO-DMT, are highly susceptible substrates for monoamine oxidase (MAO), specifically MAO-A, and hence can be greatly potentiated by monoamine oxidase inhibitors (MAOIs). An example of this is ayahuasca, in which plants containing both DMT and harmala alkaloids acting as MAOIs such as harmine and harmaline are combined. This allows DMT to become orally active and to have a much longer duration of action than usual. The 2C psychedelics, such as 2C-B, 2C-I, and 2C-E, are also substrates of both MAO-A and MAO-B, and may likewise be greatly potentiated by MAOIs. Examples of MAOIs that may potentiate psychedelics behaving as MAO-A and/or MAO-B substrates include phenelzine, tranylcypromine, isocarboxazid, moclobemide, and selegiline. Combination of MAO-substrate psychedelics with MAOIs can result in overdose and serious toxicity, including death. Other psychedelics, such as LSD, are not substrates of MAO and are not potentiated by MAOIs.
Watson was selected at pick 40 under the father–son rule in the 2002 national draft. He was initially coached as a key forward by Kevin Sheedy, who also coached his father Tim. His poor kicking by AFL standards drew criticism, and it was determined that he would be more suited to the midfield at Essendon. At the time, his weight was criticised by the media, as he was overweight for an AFL player. Over the next two seasons, he began to develop his craft in the Essendon midfield and became one of the competition's elite midfielders. Watson had a consistent season in 2009, polling ten Brownlow Medal votes and winning the Essendon best-and-fairest award, the Crichton Medal. Watson polled 335 votes in 15 of the 21 games he played in the 2009 season, with only one game missed due to an ankle injury. Watson was 46 votes ahead of runner-up Dustin Fletcher, who polled 289 votes. By the end of the 2009 season, Watson had gathered a reputation as Essendon's most important midfielder (coming first in the club's overall clearances) and improved his once-criticised kicking ability to above the standard of an AFL player. Watson was announced as Essendon captain on 21 December 2009, taking over from retired goalkicker Matthew Lloyd. Despite Essendon suffering a disappointing 2010 season, winning only seven games and finishing 14th on the AFL ladder, Watson enjoyed a successful first year as captain.
While Jung worked on his Psychology of the Unconscious: a study of the transformations and symbolisms of the libido, tensions manifested between him and Freud because of various disagreements, including those concerning the nature of libido. Jung de-emphasized the importance of sexual development and focused on the collective unconscious: the part of the unconscious that contains memories and ideas that Jung believed were inherited from ancestors. While he did think that the libido was an important source of personal growth, unlike Freud, Jung did not think that the libido alone was responsible for the formation of the core personality. In 1912, these tensions came to a peak because Jung felt severely slighted after Freud visited his colleague Ludwig Binswanger in Kreuzlingen without paying him a visit in nearby Zurich, an incident Jung referred to as "the Kreuzlingen gesture". Shortly thereafter, Jung again traveled to the US and gave the Fordham University lectures, a six-week series, which were published later in the year as Psychology of the Unconscious, and subsequently republished as Symbols of Transformation. While they contain remarks on Jung's dissenting view on the libido, they represent largely a "psychoanalytical Jung" and not the theory of analytical psychology, for which he became famous in the following decades. Nonetheless, it was their publication which, Jung declared, "cost me my friendship with Freud". Another disagreement with Freud stemmed from their differing concepts of the unconscious.

It is a laboratory-made peptide that mimics growth hormone-releasing hormone. It prompts the pituitary gland to release growth hormone and has been studied mainly in adults with HIV-associated lipodystrophy.
Administered growth hormone supplies the hormone directly, while this peptide acts upstream by prompting the pituitary to release it. The indirect route preserves pulsatile secretion and some endogenous feedback, which changes the hormone and IGF-1 profile observed after treatment.
Reductions in visceral adipose tissue appear consistently in randomized trials of the approved population. Effects on peripheral fat, cardiovascular outcomes, and use outside that population are less well established.
It is a laboratory-made peptide of forty-four amino acids whose sequence matches human growth hormone-releasing hormone, with a modified amino terminus. The modification is a short unsaturated fatty acid chain attached to the first residue. This change slows enzymatic breakdown and lengthens the time the peptide stays active in circulation.
